As the most widely used animal cell in bioengineering pharmacy, CHO cell has its advantages and disadvantages. CHO, as a host, has the following advantages in the expression of recombinant proteins:
1. It has accurate post transcriptional modification function, and the expressed protein is closest to natural protein molecules in terms of molecular structure, physical and chemical properties and biological functions;
2. It can not only adhere to the wall and grow, but also carry out suspension culture after domestication, and can withstand high shear force and osmotic pressure;
3. The expressed foreign proteins can be secreted outside the cell, and few of their own endogenous proteins are secreted outside the cell, which is convenient for the separation and purification of downstream products;
4. High density culture can be achieved in serum-free medium, which is convenient for large-scale production.
CHO cells have many advantages for the expression of foreign proteins, but there are also some shortcomings, such as: some recombinant proteins in the process of expression, their glycosylation components are unstable and difficult to purify, so more adjustments need to be made in the production process or purification process; At the same time, the culture cost of recombinant cells is relatively high, the production cycle is long, and the automation level is relatively low.
